Therapeutic Actions

Increases GABA levels in the brain, which may result in antiseizure effects. GABA is the major inhibitory neurotransmitter in the CNS; tiagabine binds to GABA reuptake sites, preventing its reuptake, and increases levels in the presynaptic neurons and the glia.


Indications



  • Adjunctive therapy in patients older than 12 yr with partial seizures


  • Unlabeled uses: Bipolar disorders, PTSD; refractory seizures in children; insomnia.



Available Forms

Tablets—2, 4, 12, 16 mg


Dosages

Adults

4 mg PO daily for 1 wk; may be increased by 4–8 mg/wk until desired response is seen; maximum dose, 56 mg/day in two to four divided doses. Usual maintenance dose is 32–56 mg PO daily.

Pediatric patients 12–18 yr

4 mg PO daily for 1 wk; may be increased to 8 mg/day in two divided doses for 1 wk; then increased by 4–8 mg/wk up to a maximum of 32 mg/day PO in two to four divided doses.

Pediatric patients younger than 12 yr

Not recommended.
