Gonadotropin-releasing hormone (GnRH) analogue
Therapeutic Actions
An LH-RH agonist that occupies pituitary gonadotropin-releasing hormone receptors and desensitizes them; inhibits gonadotropin secretion when given continuously, leading to an initial increase, then profound decrease in LH and FSH levels.
Indications
Advanced prostatic cancer—palliation, alternative to orchiectomy or estrogen therapy
Depot only: Endometriosis
Central precocious puberty
Depot only: Uterine leiomyomata
Unlabeled uses: Treatment of breast, ovarian, and endometrial cancers; infertility; prostatic hypertrophy
Contraindications and Cautions
Contraindicated with allergy to leuprolide, pregnancy, undiagnosed vaginal bleeding, lactation.

Injection—5 mg/mL; Depot—3.75, 7.5 mg; Depot-Ped—7.5, 11.25, 15 mg; 3-mo Depot—11.25, 22.5, 45 mg; 4-mo Depot—30 mg, 72 mg; 6-mo Depot—45 mg; powder for injection—7.5 mg
Dosages
Adults
Advanced prostate cancer: 1 mg/day subcutaneously; use only the syringes that come with the drug.
Depot: 7.5 mg IM monthly (every 28–33 days). Do not use needles smaller than 22 gauge.
3-mo depot: 22.5 mg IM or subcutaneously every 3 mo (84 days).
4-mo depot: 30 mg IM or subcutaneously every 4 mo.
6-mo depot: 45 mg subcutaneously every 6 mo.
Endometriosis: 3.75 mg as a single monthly IM injection or 11.25 mg IM every 3 mo. Continue for 6 mo.
Uterine leiomyomata: 3.75 mg as a single monthly IM injection for 3 mo or 11.25 mg IM once; give with concomitant iron treatment.Stay updated, free articles. Join our Telegram channel
