Therapeutic Actions
Selectively binds to serotonin (5-HT3) receptors in the CTZ, blocking the nausea and vomiting caused by the release of serotonin by mucosal cells during chemotherapy, which stimulates the CTZ and causes nausea and vomiting.
Indications
Prevention and treatment of nausea and vomiting associated with emetogenic chemotherapy and radiation
Prevention and treatment of postoperative nausea and vomiting (injection)
Unlabeled uses: Postanesthetic shivering, cancer-related pruritus
Contraindications and Cautions
Contraindicated with history of hypersensitivity, concurrent use of apomorphine.
Use cautiously with colitis, hepatic or renal impairment, pregnancy, lactation.
Available Forms
Tablets—1 mg; injection—0.1, 1 mg/mL; oral solution—1 mg/5 mL; transdermal system—3.1 mg/24 hr
Dosages
Adults and patients 2 yr and older
IV
Chemotherapy-induced nausea and vomiting: 10 mcg/kg IV over 5 min starting within 30 min of chemotherapy; only on days of chemotherapy.
