Therapeutic Actions

Potentiates GABA action by binding to GABA receptor sites. GABA is an inhibitory neurotransmitter; the exact action causing the clinical effects is not understood.


Indications



  • Adjunct treatment of seizures associated with Lennox-Gastaut syndrome in patients 2 yr and older



Available Forms

Tablets—5, 10, 20 mg


Dosages

Adults and children 2 yr and older



  • 30 kg or less: Initially, 5 mg/day PO. Increase to 5 mg PO bid starting on day 7; increase to 10 mg PO bid starting on day 14.
    Over 30 kg: Initially, 5 mg PO bid. Increase to 10 mg PO bid starting on day 7; increase to 20 mg PO bid starting on day 14.

Geriatric patients, poor CYP2C19 metabolizers, patients with mild to moderate hepatic impairment

Initially, 5 mg/day PO. Titrate according to weight at half the usual dose to a maximum of 20–40 mg/day based on weight, starting on day 21, if tolerated.
