Therapeutic Actions
Inhibits protozoal reproduction and protein synthesis. Mechanism of anti-inflammatory action in rheumatoid arthritis is not known.
Indications
Treatment of extraintestinal amebiasis
Prophylaxis and treatment of acute attacks of malaria caused by susceptible strains of Plasmodia
Unlabeled use: Treatment of rheumatoid arthritis (150 mg PO daily)
Contraindications and Cautions
Contraindicated with allergy to chloroquine and other 4-aminoquinolines; retinal or visual field changes.
Use cautiously with porphyria, psoriasis, retinal disease, hepatic disease, G6PD deficiency, alcoholism, lactation, pregnancy.

Tablets—250, 500 mg
Dosages

Adults
Amebiasis: 1 g (600 mg base)/day PO for 2 days; then 500 mg (300 mg base)/day for 2–3 wk.
Malaria: For suppression, 300 mg base (500 mg) PO once a week on the same day for 2 wk before exposure and continuing until 8 wk after exposure. For acute attack, initially, 600 mg base (1 g) PO; then 300 mg base (500 mg) 6–8 hr, 24 hr, 48 hr after the initial dose for a total dose of 1.5 g base (2.5 g) in 3 days.Stay updated, free articles.
